🔬 Overview
The LIGAN platform offers you the Illumina 5-base solution, a revolutionary multi-omic technology that combines the detection of genetic variants with comprehensive DNA methylation (5mC) analysis in a single experiment.
Leveraging our teams’ expertise in library preparation, this approach offers remarkable sensitivity, a high alignment rate and uniform coverage, making it ideal for revealing the epigenetic richness of your samples.
⚙️ General principle
The 5-base technology is based on:
A gentle and direct enzymatic conversion: unlike aggressive chemical bisulphite treatments, enzymatic conversion preserves the integrity and diversity of DNA sequences.
Bespoke manual preparation: a meticulous process carried out by our engineers, ensuring maximum flexibility and high-quality execution.
Two results in a single run: simultaneous generation of genotyping and methylation profiling, eliminating the need for two separate assays.
🌟 Benefits
Data quality: CpG site coverage up to 60 per cent higher than traditional methods (WGBS/bisulphite) and reduced GC bias.
Sample and time savings: a single preparation and a single sequencing run for two levels of biological information.

🔧 Technological infrastructure
The implementation of the 5-base solution via the LIGAN platform is underpinned by a controlled analytical workflow:
- Sample preparation:
A best-practice manual protocol (Illumina 5-Base DNA Prep) carried out by our engineers, ensuring rigorous quality control at every stage of library generation.
- High-throughput sequencing:
Illumina NovaSeq X Plus – Guaranteeing high throughput, optimal read depth and a highly competitive cost per gigabase.
- Bioinformatics analysis:
Locally hosted DRAGEN server (Illumina) – Integrated secondary processing for the decoupling of genetic variants and methylation annotations in record time.
